Mercury (Hg) is a global environmental concern due to its toxicity (especially high in methylated form) and the long-range distribution of its gaseous elemental form (GEM). Hg-contaminated areas, such as abandoned mining sites, pose intrinsic difficulties for their management and heavy monitoring costs. In these environments, plant-based solutions may play a key role in the ecosystem quality assessment and support remediation strategies, combining reliability and cost-effectiveness. In this study, we adopted a biomonitoring approach by using tree rings of four different species collected in the proximity of the mining-metallurgical area of Abbadia San Salvatore, central Italy, a major former Hg mining district whose reclamation is currently in progress. Our dendrochemical analysis was aimed at identifying the historical changes of local atmospheric Hg contamination and at singling out, for the first time in the study area, other potentially toxic elements (PTEs) associated with the past mining activity. Collected cores dated back to early as 1940 and provided the temporal patterns of atmospheric Hg emission vs the produced liquid quantities, so reconstructing the historical impact of the mining site on nearby terrestrial ecosystems and resident human population. Current GEM contamination was found about twenty times lower than that of the fully operational mine periods. From a first survey on other PTEs, thallium (Tl) and lead (Pb) appeared to be potentially associated with the mining activity, thus suggesting new working assumptions for further dendrochemical analyses and for the inclusion of Pb in human biomonitoring surveys of the Mt. Amiata area, actually not present in the control list. The results prompt a more thorough assessment by tracking for a longer time span a critical site that is an ideal open-field lab to study the ecophysiology of different tree species in relation to environmental behavior of PTEs for better-assessing wildlife and human exposures.

In this study, leaves of the evergreen holm oak Quercus ilex were used to assess airborne contamination of potentially toxic elements (PTEs) at five towns located on the slopes of the Mt. Amiata (central Italy), an area with a long history of mining and, more recently, an important district for the industrial exploitation of geothermal energy. PTE composition and covariance of washed and unwashed Q. ilex leaves of three different ages (6, 12 and 24 month-old) were used to identify atmospheric inputs of PTEs at residential areas, evaluate long-term adsorption and retention of PTEs by the leaves, thus providing an indication of potential human exposure. Moreover, the determination of foliar concentrations of major elements (C, N, S and P) allowed an assessment of the nutritional status of the investigated urban tree stands which excluded the existence of stress condition caused by air pollution or other disturbances. Results indicated that overall Pb, Cu, and Cd concentration were low in the investigated urban sites, if compared with similar studies conducted in larger Italian cities, denoting a low contribution of vehicular traffic to the atmospheric pathway. The five urban settlements were characterized by a specific profile of elements (Al, Ba, Hg and Sb) enriched in unwashed leaves, resulting from the distinct geochemical characteristics of the area and from diffuse (i.e., urban activity) and point sources of PTEs emission (i.e., brownfields, geothermal power plants). The latter sources primarily govern the distribution of Hg, whose contamination was found to be very localized close to a major abandoned mining area. Our data provided quantitative evidence of the spectrum of PTEs potentially impacting resident population and may prove useful in support of follow-up instrumental monitoring campaigns of air quality, as well as for human health and ecological risk assessments.

Lichens play an important role in the biogeochemical cycling of mercury (Hg) and are commonly used as indicators of Hg enrichment in remote and anthropogenically impacted environments. To assess their capacity for Hg uptake and accumulation, we determined the concentration of gaseous elemental mercury (GEM) in air and the concentration of total Hg (THg) in transplanted thalli of two lichen species. Lichen transplants and passive air samplers (PASs) were concurrently deployed, side by side, at 10 sites within an abandoned mining area, characterized by large gradients in atmospheric Hg contamination. Highly variable time-weighted GEM concentrations determined by the PASs, ranging from 17 to 4,200 ng/m3, were mirrored by generally high Hg concentrations in transplanted thalli of both Xanthoria parietina (174-8,800 ng/g) and Evernia prunastri (143-5,500 ng/g). Hg concentrations in the two species co-varied linearly indicating about 60% greater Hg accumulation in X. parietina than in E. prunastri. Whereas Hg uptake in the fruticose E. prunastri increased linearly with GEM, a power law equation with a fractional exponent described the uptake in the foliose X. parietina. Extrapolating the relationships observed here to higher GEM levels yielded concentrations in lichen that agree very well with those measured in an earlier fumigation experiment performed under laboratory-controlled conditions. The uptake model of X. parietina was further verified by correctly estimating GEM concentrations from the THg measured in autochthonous thalli collected from the urban area adjacent to the mine site. Passive sampling can effectively provide time-weighted data of suitable spatial resolution to quantitatively describe GEM assimilation by lichens. Therefore, the combined use of passive sampling and lichen transplants can contribute to a more comprehensive understanding of the role of lichens, and potentially also of other cryptogams, in the deposition of atmospheric Hg to terrestrial ecosystems.

Bio-stimulants are showing growing success and are gradually replacing synthetic fertilizers in agriculture. Wood distillate (WD), also known as wood vinegar or pyroligneous acid, is a by-product of biomass pyrolysis and is increasingly used as a bio-stimulant for crop production. This study investigated whether weekly foliar applications of 0.25% and 0.50% WD have a differential effect on the chlorophyll and sugar content as well as biomass production in lettuce (Lactuca sativa L.). Moreover, the additional beneficial effect from the addition of corroborants of plant origin such as 3% soy lecithin and 5% flavonoid-rich wood glycolic extract to WD (BF) was investigated. Moreover, the possible toxicological concern from some potentially toxic elements (PTEs), namely Cd, Cu, Fe, Pb, and Zn, which may be abundant in WD was verified. After four weeks, we found that 0.25% WD not only increases lettuce biomass, which has an economic value, but also has beneficial effects on other qualitative parameters such as sugars and total sweetness. On the other hand, the use of 0.5% WD decreased the content of soluble sugars, suggesting a hormetic-type effect. We did not find evidence of further beneficial effects from the addition to WD of plant-derived corroborants, nor of any enrichment in the content of the investigated PTEs.

This study investigated the dynamics of the accumulation and release of Hg2+ in lichens, using Evernia prunastri (L.) Ach. as a model species. Thalli were incubated with solutions containing 1, 10, and 100 µM Hg2+ and then exposed for 1, 2, 3, 6, 12, 18, and 24 months at the Botanical Garden of the University of Siena (a location free from local Hg sources). Lichen samples accumulated Hg proportionally to the exposure concentration, and after the exposure, reductions over time were evident, already starting from 1-2 months. After 24 months, samples released 72-74 (healthy thalli) to 94% (unhealthy thalli) of the accumulated Hg, but control values of untreated samples were never reached. Depending on the Hg content after the exposure, stable decreased concentrations were reached after 6-24 months. The results of this study highlight the ability of the lichen E. prunastri to reflect rapidly increasing environmental Hg concentrations, as well as to indicate an ameliorated situation (e.g., the closure of an Hg source). However, we have found evidence that an acute pollution episode can influence the content of Hg in lichens for several years.

In this study, the release of Cu2+ and Zn2+ was investigated and modeled in the epiphytic lichen Evernia prunastri. Samples were incubated with solutions containing these metals at ecologically relevant concentrations (10 and 100 µM) and then transplanted to a remote area and retrieved after 1, 2, 3, 6, 12, and 18 months. The results showed that, after 12 months, all samples faced similar metal reductions of ca. 80-85%, but after this period, all the involved processes seem to be no longer capable of generating further reductions. These results suggest that the lichen E. prunastri can provide information about environmental improvements after exposure to high or very high pollution levels in a relatively short period of time.

Abandoned mine sites are a cause of great environmental concern, being potential sources of toxic elements for adjacent aquatic ecosystems with intrinsic difficulties for their management (i.e. episodic nature of pollution, technical difficulties and high costs of monitoring, remoteness). Aquatic macrophytes can find effective application in these situations, providing cost-effective data for instream water quality assessment. In this study, native and transplanted specimens of the aquatic moss Platyhypnidium riparioides were used to evaluate metal(loid) contamination in a river receiving multiple acidic and metalliferous drainages from sulphide mineralized areas and derelict mines. Analysis of native P. riparioides thalli was used to identify, in the upland course of the river, the pattern of contamination (As, Cd, Cu, Pb and Zn) which was related to the geo-environmental features of the watershed and the nearby historical mining areas. Attenuation of metal(loid) availability in the lowland river, apparently due to eco-hydrological and physic-chemical processes, was also highlighted by spatial trends of concentrations data of native and transplanted moss. The latter, deployed for 21 days at specific stretches of the river and in a tributary hydrologically connected with a dismissed mine, supported the identification of point sources (i.e. mine effluents, metallurgical waste piles amassed on the banks of the river) and the reckoning of their quantitative impact on different segments of the watercourse. By exploring multi-elemental and native-to-transplant relationships, differences in metal(loid) accumulative capacities were recognized between sampled thalli and exposed moss bags in relation to the severity of the contamination. The observed discrepancy in the accumulation of As, Fe, Ni and Pb in highly contaminated areas between native and transplanted moss of P. riparioides raises questions on the possible competing mechanisms of element uptake and retention. These findings prompt studies to discern possible limitations of the transplanting moss technique under extreme stream-quality conditions.

Treatments of crops with additives to increase their productivity may pose environmental risks and induce negative effects also on non-target organisms. In this study, we investigated the potential effect of chestnut wood distillate (pyroligneous acid) used in agriculture, on the accumulation of trace elements in aquatic plants. As a model species, the common water fern Azolla filiculoides Lam. was selected, being often used also in phytoremediation processes. The content of selected elements of toxicological concern (As, Ba, Cd, Cu, Fe, Mn, Ni, Pb, Zn) was assessed in the fern after short-term treatments (1-3 days) over a range of wood distillate concentrations 1:300 (3.33 mL/L), 1:500 (2.00 mL/L), 1:700 (1.43 mL/L). A statistically significant accumulation of Cd, Cu, Mn, Pb, Zn (1:700) and Pb (1:300) was recorded after three days of incubation, despite the concentrations remained overall low. Using treatment vs. control ratios, a trend of increasing temporal uptake was detected for As, Ba, Fe, Mn, Pb (1:700); Mn, Pb (1:500), and only Pb at 1:300. The results suggested that, under the experimental conditions, element uptake is positively influenced by time and negatively by increasing concentrations of wood distillate, likely due to the acidification of the medium. On the whole, the element concentrations measured in A. filiculoides were low and did not pose any toxicological concern.

This study compared the ability of the lichen Evernia prunastri, chitin and chitosan to take up Cu2+ and Zn2+. It was hypothesized that chitin and chitosan have an accumulation capacity comparable to the lichen, so that these biopolymers could replace the use of E. prunastri for effective biomonitoring of Cu and Zn air pollution. Samples of the lichen E. prunastri, as well as chitin (from shrimps) and chitosan (from crabs), were incubated with Cu and Zn solutions at concentrations of 0 (control), 10, 25, 50, 75, and 100 µM and analyzed by Inductively Coupled Plasma Mass Spectrometry (ICP-MS). Metal concentrations accumulated by lichen, chitin and chitosan samples were strongly and linearly correlated with the concentrations in the treatment solutions. The lichen always showed significantly higher accumulation values compared to chitin and chitosan, which showed similar accumulation features. The outcomes of this study confirmed the great effectiveness of the lichen Evernia prunastri for environmental biomonitoring and showed that chitin and chitosan have a lower accumulation capacity, thus suggesting that although these biopolymers have the potential for replacing E. prunastri in polluted areas, their suitability may be limited in areas with intermediate or low pollution levels.

Wood distillate (pyroligneous acid) can be successfully applied in agriculture to increase crop quality and productivity with a lower risk for the environment respect to synthetic chemical herbicides, pesticides or fertilizers. However, the effects of wood distillate on the environment and biota are still under investigation, depending on biological attributes of potentially influenced organisms. The potential toxicological effects of wood distillate on sensitive non-target organisms, lichens and mosses, are studied for the first time. The physiological parameters (chlorophyll a fluorescence emission FV/FM and PI(ABS), chlorophyll content, spectral reflectance, antioxidant power, and dehydrogenase activity) and eventual bioaccumulation of selected elements (As, Ba, Cd, Cr, Cu, Fe, Ni, Pb, Zn) were investigated in the lichen Xanthoria parietina and the moss Hypnum cupressiforme after short-term treatments over a range of wood distillate solutions (1:300, 1:500, 1:700) to detect potential early stress responses. Overall, the lichen did not show changes after the treatments, while in the moss wood distillate caused only modest alterations in FV/FM and PI(ABS) and progressive increasing of antioxidant activity according to the dose supplied. The bioaccumulation of toxic elements was low and did not show any pattern of uptake with increasing concentrations of wood distillate.

Tracing emission sources and transformations of atmospheric mercury with Hg stable isotopes depends on the ability to collect amounts sufficient for reliable quantification. Commonly employed active sampling methods require power and long pumping times, which limits the ability to deploy in remote locations and at high spatial resolution and can lead to compromised traps. In order to overcome these limitations, we conducted field and laboratory experiments to assess the preservation of isotopic composition during sampling of gaseous elemental mercury (GEM) with a passive air sampler (PAS) that uses a sulfur-impregnated carbon sorbent and a diffusive barrier. Whereas no mass independent fractionation (MIF) was observed during sampling, the mass dependent fractionation (MDF, δ202Hg) of GEM taken up by the PAS was lower than that of actively pumped samples by 1.14 ± 0.24‰ (2SD). Because the MDF offset was consistent across field studies and laboratory experiments conducted at 5, 20, and 30 °C, the PAS can be used for reliable isotopic characterization of GEM (±0.3‰ for MDF, ±0.05‰ for MIF, 2SD). The MDF offset occurred more during the sorption of GEM rather than during diffusion. PAS field deployments confirm the ability to record differences in the isotopic composition of GEM (i) with distance from point sources and (ii) sampled at different background locations globally.

The worldwide growing interest in traditional medicines, including herbal medicines and herbal dietary supplements, has recently been accompanied by concerns on quality and safety of this type of health care. The content of nutritional and potentially toxic elements in medicinal plants is of paramount interest as it may vary remarkably according to different environmental and ecophysiological factors. In this study, the concentrations of essential and non-essential trace elements-Co, Cr, Cu, Ni, Sr, and Zn-were determined in the roots and aerial parts of the worldwide distributed and economically important medicinal herb Hypericum perforatum L. (St. John's wort) and in its growing substrate. Most of the analyzed trace elements varied considerably in the plant parts according to edaphic conditions and soil geochemistry. However, uptake and retention in H. perforatum compartments of Co, Cr, and Ni, which markedly differentiated the investigated soils, were controlled by excluding mechanisms of the plant. Despite this, the Ni concentrations in the aerial parts, commonly used in herbal preparations, of H. perforatum plants from serpentine soils were not insignificant in relation to eventual human consumption. Good practice to assure the herbal product quality of H. perforatum collected from the wild cannot ignore the thorough understanding of the geolithological and geochemical features of the harvesting areas.

This study evaluated the biological effects of highly polluted freshwater environment (Regi Lagni channels, S Italy) on the aquatic moss Leptodictyum riparium, exposed in bags at three sites representative of different environmental conditions and characterized by different heavy metal burdens. Bioaccumulation, ultrastructural alterations, Reactive Oxygen Species (ROS) production, antioxidant enzymes activity and DNA damage were assessed. To better evaluate the biological response of the moss species to heavy metals, the same biological parameters were assessed also in L. riparium samples cultured in vitro using metal mixtures at the same concentrations as measured at the 3 field exposure sites. Heavy metals were accumulated into the moss tissues causing severe ultra-structural damages at higher concentration case studies, and the ROS production as well as the activity of the enzyme followed a concentration-dependent increase. However, the DNA damage trend suggested a threshold effect that changed between field and in vitro experiment. The enrichment factor suggests that the concentration in the most polluted site is close to the upper limit of L. riparium to accumulate metals. Overall, combining measures of the morpho-functional traits at different level contribute to improving the knowledge about the tolerance of L. riparium to heavy metal stress, suggesting that this moss could be suitable for biomonitoring activity in field conditions.

This study evaluates the effects of toxic metal pollution in the highly contaminated Sarno River (South Italy), by using the aquatic moss Leptodictyum riparium in bags at 3 representative sites of the river. Biological effects were assessed by metal bioaccumulation, ultrastructural changes, oxidative stress, as Reactive Oxygen Species (ROS) production and Glutathione S-transferase (GST) activity, as well as Heat Shock Proteins 70 (HSP70s) induction. The results showed that L. riparium is a valuable bioindicator for toxic metal pollution of water ecosystem, accumulating different amounts of toxic metals from the aquatic environment. Toxic metal pollution caused severe ultrastructural damage, as well as increased ROS production and induction of GST and HSP70s, in the samples exposed at the polluted sites. To assess the role and the effect of toxic metals on L. riparium, were also cultured in vitro with Cd, Cr, Cu, Fe, Ni, Pb, Zn at the same concentrations as measured at the 3 sites. Ultrastructure, ROS, GST, and HSP70s resulted severely affected by toxic metals. Based on our findings, we confirm L. riparium as a model organism in freshwater biomonitoring surveys, and GST and HSP70s as promising biomarkers of metal toxicity.

The competitive behavior of divalent heavy metals (Cd, Cr, Pb, Zn) during cation uptake was investigated in the foliose lichen Xanthoria parietina. Lichen thalli were incubated with solutions containing 10 and 100 µM of CdCl2, CuCl2, and ZnCl2 as well as 5 and 50 µM of Pb(NO3)2, tested individually and in combination (Cd2++Cu2++Pb2++Zn2+). The analysis of molar concentrations suggests that a competition between cations for binding sites in X. parietina does exist. The decrease in net uptake between single and mixed solutions ranged between 14 and 29% at the lowest concentration and between 38 and 68% at the highest concentration. Furthermore, the uptake was proportionally lower for richer solutions. Each metal may behave differently when uptook: some (toxic elements) are preferentially stored at extracellular level (Cd, Pb), while others (micro-nutrients) are also present at intracellular level (Cu and Zn). The proportion between extracellular and total content changed for those elements accumulated also at intracellular level (Cu and Zn), while for Cd and Pb almost all the uptake occurred by passive mechanisms mainly at extracellular binding sites. The competition between metals for binding sites in the lichen surface entails that bioaccumulation data might result in an underestimation of some element levels measured in biomonitoring studies.

The liverwort Lunularia cruciata, known for being a species tolerant to pollution able to colonize urban areas, was collected in the town of Acerra (South Italy) to investigate the biological effects of air pollution in one of the three vertices of the so-called Italian Triangle of Death. The ultrastructural damages observed by transmission electron microscopy in specimens collected in Acerra were compared with samples collected in the city center of Naples and in a small rural site far from sources of air pollution (Riccia, Molise, Southern Italy). The biological response chain to air pollution was investigated considering vitality, photosynthetic efficiency, heat shock protein 70 (Hsp70) induction and gene expression levels, and chlorophyll degradation and related ultrastructural alterations. Particularly, a significant increment in Hsp70 expression and occurrence, and modifications in the chloroplasts' ultrastructure can be strictly related to the environmental pollution conditions in the three sites. The results could be interpreted in relation to the use of these parameters as biomarkers for environmental pollution.

We investigated the influence of titanium dioxide nanoparticles (nano-TiO2) on the response to cadmium in the gills of the marine mussel Mytilus galloprovincialis in terms of accumulation and toxicity. Mussels were in vivo exposed to nano-TiO2, CdCl2, alone and in combination. Several cellular biomarkers were investigated in gills: ABC transport proteins and metallothioneins at gene/protein (abcb1, abcc-like and mt-20) and functional level, GST activity, NO production and DNA damage (Comet assay). Accumulation of total Cd and titanium in gills as in whole soft tissue was also investigated. Significant responses to Cd exposure were observed in mussel gills as up-regulation of abcb1 and mt-20 gene transcription, increases in total MT content, P-gp efflux and GST activity, DNA damage and NO production. Nano-TiO2 alone increased P-gp efflux activity and NO production. When combined with Cd, nano-TiO2 reduced the metal-induced effects by significantly lowering abcb1 gene transcription, GST activity, and DNA damage, whereas, additive effects were observed on NO production. A lower concentration of Cd was observed in the gills upon co-exposure, whereas, Ti levels were unaffected. A competitive effect in uptake/accumulation of nano-TiO2 and Cd seems to occur in gills. A confirmation is given by the observed absence of adsorption of Cd onto nano-TiO2 in sea water media.

Heavy metals (HMs) and polycyclic aromatic hydrocarbons (PAHs) were analysed in topsoil and in Quercus ilex L. leaves from holm oak woodlands located along urbanization gradients (urban, periurban and extraurban sites) in two Italian regions (Campania and Tuscany). In each sampling site, the metal bioavailability factors (MBFs) and the pollutant bioaccumulation factors (BAFs) were calculated to estimate the fraction of each total metal concentration in soil potentially available to root uptake and to know the fate of both HMs and PAHs in the plant-soil system. In general, the results indicated a low atmospheric deposition of pollutants and, in some cases, leaves and soils accumulated higher HM and PAH concentrations in the most urbanized areas. Correlation analyses showed that in each sampling site topsoil and leaves were exposed to the same atmospheric inputs of HMs and PAHs, although to a different extent. Notwithstanding the MBFs and BAFs differed between the two regions for the most HMs, they did not show a clear pattern in relation to the urbanization gradient. However, some information could be deduced by these ratios: the high BAF calculated for Mn shows that the foliar uptake plays an important role in accumulation of this metal. Moreover, for PAHs the different values of BAFs among low and high molecular weight compounds suggest their different fate, the first accumulated in leaves by stomata and the latter preferentially deposited on topsoil.

We determined genetic variation and metal and polycyclic aromatic hydrocarbon concentrations in Leptodon smithii moss collected in holm oak stands at cities, outskirts and remote areas of Campania and Tuscany (Italy) to investigate if anthropogenic pressure (pollutant emissions and land use change) affects moss genetic richness. In both regions, metal and polycyclic aromatic hydrocarbon concentrations reflected the trend urban>outskirts>remote areas, excepting Tuscany remote site. In both regions, the moss gene diversity increased from urban to remote areas. The findings suggest the extent and the fragmentation of urban green areas, as drivers of moss genetic richness.
